Interaction between microRNA-493-5p and CCL19 dictates migration, invasion and progression of non-small cell lung cancer
Abstract
The chemokine, CCL19 can promote lung cancer invasion and metastasis by binding to its receptor CCR7, but its specific mechanism is still unclear. We performed - miRNA microarray analysis to assess- miRNA expression in A549 cells treated with or without CCL19, and miR-493-5p was chosen for further study. Then, the role of miR-493-5p -in migration, invasion and EMT -were evaluated. miR-493-5p was significantly downregulated in A549 cells after CCL19 treatment. miR-493-5p -attenuated the downregulation of E-cadherin expression -after CCL19 treatment and -upregulated N-cadherin and Snail expression. Moreover, miR-493-5p overexpression -attenuated the effect of CCL19 on promoting migration and invasion as well as c-Met and Akt expression in A549 cells. miR-493-5p expression was downregulatedin lung cancer tissues, especially those from patients with lymph node or vascular metastasis. CCL19 can downregulate the expression of miR-493-5p and abolish the inhibitory effect of miR-493-5p on c-Met, which may activate the AKT signaling pathway, promote epithelial-mesenchymal transition, and increase the migration and invasionof A549 cells.
